Did this drive a few months ago (in an my uncle's Alfa 159).
Stayed in Apollo Bay, then spent the next day in the Otways (awesome), then the next day continued to Port Fairy and did the twelve Apostles. On the way back went up through the Grampians. Beautiful.
I can probably find the names of the actual places we stayed if you want. We LOVED "The Victoria Apartments" in Port Fairy, stayed there for only one night though. I really liked Port Fairy. Liked getting out and hiking in the Grampians and Otway the most. During Holidays the road I'm sure gets very crowded. Lots of tour buses near the twelve apostles.
In terms of "giving it some" on the road, there were some fun places to drive along the way with nice twisties, but for most of it it was fun enough just looking out of the window. I'll throw some pics up in a minute.
I'm also a huge "nature" guy and was totally nerding out about all the parrots and tree-ferns. Not to mention all the badass cars they never imported to the states.
