If you were driving it from a long lay up it could be something like a sticking rear brake shoe/disc pad, some times after driving in heavy rain if you put your car away and don't use it for a while, the first time you use it the pads/brake shoes could be stuck to the drums/discs.
I would check this before stripping the diff.
