I distinctly remember having to load the drivers separately. I messed with a Blutooth connection between the computer and my mobile phone sitting in the garage. Got it to work after a while. There should be a thread about this, but I can't find it.

The drivers which are particular to S&S are loaded onto the computer with the latest version of Protune II. However, unless the computer recognises a connection to the module the drivers will not load into the registry. No internet connection is required.
Ive tried Windows 7, Windows 10 and even a Mac (running a hypervisor) in all cases, once the ECM is powered and recognised the drivers load.
Rebooting may be required once the drivers are loaded. Try a different USB lead, make sure ignition is on

Grockle, Thanks for the advice. I have actually tried everything you have suggested. I am running Windows 10 on a new MS Surface Pro 4. I am using the mini USB cable that came with my GoPro. When I loaded the S&S software I never received a message asking me if I wanted to load the drivers anyway. I connected to the ECU to the USB, powered on the ignition, and nothing happened. The drivers didn't load. I tried it multiple times, including uninstalling ProTune II, and reinstalling it. Rebooting, etc. I even tried watching this video, but I still couldn't get the connection to work. I gave up after about 45 mins of trying. https://www.youtube.com/watch?v=A_BvpG-s30sI am feeling frustrated not knowing what is causing my check engine light. It doesn't seem like anything is wrong, but it would be good to know why the light is on. Also, I noticed that my engine break-in period is taking rather long! I have about 8 hours of use with 185 miles on the odometer, and my rev limiter still is kicking in around 4600rpm. When does the rev limiter go up??? It's hard to find nice days to drive the M3W, but I am anxious to get the rev limiter off! Thanks guys! Hi I have done a bit of looking at the Ecu via ProTune. Using Win 10 at the moment.
My only suggestion would be to try a different or all the available USB ports on the computer. I found that there was success with one port, but not others. Why? no bloody idea.

some of the most possibles reasons 1/ The cable is only charge and no data 2/ your USB port is out of date (USB 1.0 , 2.0) when the ECU need more recent one 3/ ECU need cable support higher power and your cable is Weak (eg: ECU need 2.0 amp but cable only support 1.5amp) 4/ your ECU can't be read without specific software
as you can see most of the reason turn around the ECU
PS: As ECU is write in SSD/Flash memory the question of power on/off should not enter for start to read it (opposite as HD powered disk)

Polar, Nigels suggestion about using an alternative USB port is quite valid and worth trying. You seem to have all the other bases covered and I can confirm the GoPro cable works fine.
Im wondering if there may be a conflict on some PC's stopping detection of the ECM.
As a next step I would try to borrow a machine, it doesn't have to be high spec, Protune II doesn't need anything fancy.

I believe S&S had the drivers on their web site for separate downloading when I did it, but that was with a Win 8.1 machine = maybe up to three years ago. ProTune II still worked for me after upgrading the computer to Win 10.
Have you tried asking S&S for the drivers?

These are all really great suggestions. Thanks so much guys for all of your input and advice.
So I tried the following…
- New mini USB cable bought on Amazon. - Uninstalled and reinstalled ProTune2 software - Made sure I had full admin rights to my windows 10 computer - Try to find the drivers separately on the S&S website, but could not locate them
Unfortunately the combination of all these new things that I have tried has not worked. Not quite sure what to do at this point but it's the off-season right now, and the M3W is not saying much use. I will likely wait till the spring, and take it in to the dealer for its first service.
Any input on the break-in period?

There is a video on Youtube about how to install ProTune II, from S&S. Have you seen it?
Will try to go through my old computer, as well as the newest one, to look for the drivers.
